Description: MC192-Saporin is an immunotoxin comprising of the monoclonal antibody MC192 against rat p75NTR , the nerve growth factor receptor, chemically conjugated to the ribosome-inactivating toxin, saporin, from saponaria officinalis. Naked saporin is incapable of entering the cells due to the apparent lack of ligand. Upon specific binding via MC192 to the cells expressing p75NTR, the toxin is transverse the cell membrane leading to lesion of neurochemically defined neuronal population. The target of MC192-Saporin immunotoxin is p75NTR-expressing cells including Cholinergic neurons of the basal forebrain, Cerebellar Purkinje cells, Medial septum, Diagonal band of Broca, Nucleus basalis of Meynert and some tumour cells. This immunotoxin has been used in the study of learning and memory. The immunotoxin may be administrated via an intraventicular injection.
Handling: Only qualified personnel should handle this product. Extreme precaution must be employed when handling the immunotoxin. Wear lab coat, gloves and safety glasses. The toxin must not come into contact with open wounds. Practice good laboratory technique to avoid auto-exposure/auto-injection! Wash thoroughly any part of the body exposed to the immunotoxin. Contaminated materials should be exposed to 300 mM NaOH or autoclaved.
Applications: 1. To specifically target and eliminate rat cells expressing p75NTR. MC192-Saporin has been employed to selectively lesion cholinergic neurons of basal forebrain to create an animal model to study Alzheimer's disease. 2. To be used as a model for gene delivery into neurons.
Form: Lyophilised
Reconstitution: MC192-Saporin is delivered to you in a lyophilised form from PBS. Pulse-centrifuge to collect the immunotoxin in the bottom of the tube. Resuspend in a desired volume. Further dilution may be made in PBS. Gently triturate the sample.
Storage: Aliquot and keep the undiluted stock at -80°C for long-term storage. Avoid repeated thaw-freezing as this may result in inactivation of the immunotoxin. We have not however, observed any significant loss of activity of the stock upon storage at 4°C for up to 6 months.
