| Description: |
MDM2 is a nuclear phosphoprotein that binds and inhibits transactivation by p53 as part of an autoregulatory negative feedback loop. Overexpression of MDM2 can result in excessive inactivation of p53 thereby diminishing its tumor suppressor function. MDM2 functions as a ubiquitin ligase E3 permitting the nuclear export of p53 and its proteasome-mediated proteolysis. MDM2 also affects the cell cycle, apoptosis and tumorigenesis through interactions with other proteins such as p73, ARF(P14), retinoblastoma 1 and ribosomal protein L5. At least 11 isoforms of MDM2 are produced by alternative splicing. |

Immunohistochemistry (IHC) and Western Blotting (WB). A concentration of 0.5-1.0 µg/ml is recommended for WB. Human MDM2 (isoform MDM2) has a predicted length of 491 residues and MW of 55 kDa. A concentration of 1.0-2.0 µg/ml is recommended to detect MDM2 in formalin fixed and paraffin embedded tissues as well as formalin/acetone fixed frozen tissues. |
