Description TR-604-TAG is a luminescent tag that can be used as a conjugation reagent to azide-activated molecules for detection in cells or tissues. Azide-Alkyne cycloaddition "Click" chemistry is utilized to couple the terminal alkyne to an azide-functionalised molecule of interest, in the presence of catalytic copper. The reactions require simple or no workup or purification of the product. In unconjugated form, TR-604-TAG is a cell permeable fluorophore with large stoke shift and excellent resistance to photobleaching which makes it an ideal for fluorescence imaging and compatible with other commercially available fluorescent stains. Applications Sample types are user-defined. TR-604-TAG is a luminescent probe for copper-facilitated "Click" reactions with azide-activated molecules. Refer to the product manual for correct handling. In unconjugated form, TR-604-TAG is cell-permeable and can be used for fluorescence imaging (Ex/Em: 405 nm / 570 nm).
Specificity "Click" chemistry reagent for conjugation to azide-acitvated target molecule. Unconjugated TR-604-TAG is cell-permeable and accumulate intracellularly.
Reconstitution Spin vial briefly before opening. TR-604-TAG is soluble in both CH2Cl2 and DMF. It is insoluble in H2O. The choice of solvent system depends on the solubility of the azide used. If an organic soluble azide is used, then there is no need to include H2O in the initial reaction mixture. However, H2O will still be required in order to solubilise sodium ascorbate and CuSO4 x 5 H2O prior to addition. Refer to the product manual for coupling of both soluble and non-soluble azide derivates or azide-functionalized molecules.
Storage Product is shipped at room temperature. Store lyophilised product at room temperature, properly sealed and protected from light. Reconstituted dye should be used immediately and not stored for later use.
Expiry Date Lyophilised product is stable for 6 months at room temperature if protected from light.
