Description TrkA is a member of the neurotrophic tyrosine kinase receptor family. It is a membrane-bound receptor that upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. TrkA is required for high-affinity binding to nerve growth factor (NGF), neurotrophin-3 and neurotrophin-4/5 but not brain-derived neurotrophic factor (BDNF). TrkA leads to cell differentiations and may play a role in specifying sensory neuron subtypes. It has a crucial role in the development and function of the nociceptive reception system as well as establishment of thermal regulation via sweating. SUBUNIT: Exists in a dynamic equilibrium between monomeric (low affinity) and dimeric (high affinity) structures. SUBCELLULAR LOCATION: TrkA is a heavily glycosylated transmembrane protein and contains 13 potential N-linked glycosylation sites. The TrkA-Fc chimera has N-linked and may have O-linked oligosaccharides.

Produced in Human - A DNA sequence encoding the signal peptide and extracellular domain of human TrkA (amino acids 1- 413) was fused to the Fc region of human IgG1 (amino acids 104-330). The chimeric protein was expressed in modified human 293 cells.
Molecular Weight The TrkA-Fc chimera migrates as a broad band between 90 and 120 kDa in SDS-PAGE under reducing conditions due to post-translational modifications, in particular glycosylation. The unmodified TrkA-Fc chimera that has a predicted molecular mass of 70.2 kDa and predicted pI of 5.8. Under non-reducing conditions, TrkA-Fc migrates as a broad band between 180 and 240 kDa demonstrating the existence of this protein as a dimer. A second, less intense band is observed at 140 kDa under these SDS-PAGE conditions, suggesting the presence of non-glycosylated TrkA-Fc isoforms.

Reconstitution Spin vial briefly before opening. Reconstitute in 100 uL sterile PBS, pH 7.4 to give a concentration of 0.5 mg/mL.
Storage Lyophilized product should be stored at 2 to 8C. Following reconstitution, short-term storage at 2-8C is recommended and longer-term storage of aliquots at -18 to -20C. Repeated freeze/thawing is not recommended.
