Ubiquitin-like modifier involved in intra-Golgi traffic. Modulates intra-Golgi transport through coupling between NSF activity and SNAREs activation. It first stimulates the ATPase activity of NSF which in turn stimulates the association with GOSR1 (By similarity). Involved in autophagy. Plays a role in mitophagy which contributes to regulate mitochondrial quantity and quality by eliminating the mitochondria to a basal level to fulfill cellular energy requirements and preventing excess ROS production. Whereas LC3s are involved in elongation of the phagophore membrane, the GABARAP/GATE-16 subfamily is essential for a later stage in autophagosome maturation. SUBUNIT: Monomer. SUBCELLULAR LOCATION: Golgi apparatus. TISSUE SPECIFICITY: Ubiquitous. Expressed at high levels in the brain, heart, prostate, ovary, spleen and skeletal muscle. Expressed at very low levels in lung, thymus and small intestine. SIMILARITY: Belongs to the MAP1 LC3 family. ESTIMATED MOLECULAR WEIGHT: 13.667kDa

Reconstitution Spin vial briefly before opening. Reconstitute in 100 L of sterile water. Centrifuge to remove any insoluble material. Final buffer contains no preservatives.
Storage After reconstitution divide into aliquots and store at -20C for a higher stability. Antibody contains no preservatives. Store at 4C with an appropriate antibacterial agent. Use sterile methods. Highest purity Glycerol (1:1) may be added for an additional stability when stored at refrigerated or freezing temperatures. Avoid repetitive freeze/thaw cycles.
Expiry Date 12 months after purchase if unopened.

Mouse monoclonal antibody to GABARAP L2 (16-26), [Clone BS392]: IgG Fixing and Permeabilization of cells: Absolute methanol (10 minutes in ice) and 0.1% Tween-20 in PBS, Blocking: 1% BSA, Primary antibody: Mouse Monoclonal antibody to GABARAP L2 (cat # M-1728-100, 2μg per ~10^6 cells) for 30 minutes at room temperature, Secondary antibody: Goat anti-mouse PE labeled secondary antibody (1:100 fold dilution) with incubation for 20 minutes in dark at room temperature. Non-specific Control IgG, clone X63 (cat # M-1249-200) was used as negative control under same conditions (black dashed). Flow cytometry data and results were generated using Orflo MoxiflowTM instrument and protocols. The data demonstrates specific staining of GABARAP L2 expressed in human neuronal cell line SHSY-5Y using cat # M-1728-100.
