Mouse monoclonal antibody to slow skeletal Myosin [NOQ7.5.4D]: ascites

$327.00USD

Catalogue No. M-1830-100
Description Myosins are actin-based motor molecules with ATPase activity essential for muscle contraction. Forms regular bipolar thick filaments that, together with actin thin filaments, constitute the fundamental contractile unit of skeletal and cardiac muscle (Ref: uniprot.org).
Batch No. See product label
Unit size 100 uL
Antigen Human skeletal muscle myosin purified from myofibrils
Antibody Type Mouse monoclonal
Isotype IgG1
Clone NOQ7.5.4D
Other Names Myosin-7; Myosin heavy chain 7; Myosin heavy chain slow isoform; MyHC-slow; Myosin heavy chain, cardiac muscle beta isoform; MyHC-beta
Accession P12883 MYH7_HUMAN
Produced in Mouse
Purity Unpurified mouse ascites.
Applications Immunohistochemistry (IHC) and Western Blotting (WB). A concentration of 0.5-2.0 ug/mL is recommended for WB. A concentration of 1.0-2.0 ug/mL is recommended to detect the protein in paraffin embedded sections by IHC. Heat-mediated antigen retrieval is required. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.
Specificity The specificity of this antibody has been confirmed by WB and IHC against the antigen.
Species Against Human
Antibody Against Slow skeletal myosin
Cross-reactivity Mouse; Rat; Rabbit
Blast it If you wish to use this product in a species other than those specified here and want to determine the amino acid sequence homology between the immunogen and the corresponding protein in that species, simply copy the amino acid sequence for this immunogen and test it on the BLAST database that you can access through the link below. Please note that antibodies raised against synthetic peptides are quite often very specific for that peptide. What this means in practise is that a single amino acid difference may be enough to restrict the specificity to a particular molecule. The BLAST database can also be used to compare any amino acid sequence between species and between protein. The full amino acid sequence of the target protein can be found by following the link in the Accession field. You can then search any amino acid sequence in the target protein using the BLAST database. When selecting a sequence to search, please choose carefully, as the sequence you choose may be a precursor rather than the mature protein for example.
Form Lyophilized from mouse ascites fluid containing 1.2% sodium acetate, 2 mg BSA, and 0.01 mg NaN3 as preservative.
Reconstitution Reconstitute in 100 uL of sterile water. Centrifuge to remove any insoluble material.
Storage Store lyophilized antibody at 2-8C. After reconstitution keep aliquots at -20C for a higher stability, or at 2-8C for one month. Avoid repetitive freeze/thaw cycles.
Expiry Date 12 months after purchase

Mouse monoclonal antibody to slow skeletal Myosin [NOQ7.5.4D]: ascites Western blot analysis of Myosin (Skeletal, Slow) using anti-Myosin (Skeletal, Slow) antibody (Catalog # M-1830-100). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: mouse skeletal muscle tissue lysates. Lane 2: rat skeletal muscle tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. The membrane was blocked with 5% Non-fat Milk/TBS for 1.5 hour at room temperature. The membrane was incubated with mouse anti- Myosin (Skeletal, Slow) antibody (Catalog # M-1830-100) at 0.5 ug/mL overnight at 4C, then washed with TBS-0.1% Tween20 3 times with 5 minutes each and probed with a goat anti-mouse IgG-HRP secondary antibody at a dilution of 1:10,000 for 1.5 hour at room temperature. A specific band was detected for Myosin (Skeletal, Slow) at approximately 200-220 kDa. The expected band size for Myosin (Skeletal, Slow) is at 220 kDa.
