Product DescriptiongoogleThe rat Matrix metalloproteinase-2 (MMP-2) Kit is a sandwich ELISA. The capture antibody is a monoclonal rat MMP-2 antibody pre-coated onto the 96-well strip plates provided in the kit. Test samples and standards of known MMP-2 concentration are added to these wells and allowed to complex with the bound MMP-2 antibody. A biotinylated anti-rat MMP-2 antibody is then added. This detection antibody binds to the antigen thus completing the sandwich. After washing, an enzyme Avidin-Biotin-Peroxidase complex (ABC) is added which binds to the second antibody. The peroxidase substrate TMB is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of MMP-2 present in the samples. The purpose of this kit is the in-vitro quantitative determination of rat MMP-2 in samples such as culture supernatant, serum and plasma (Heparin). This kit has been configured for research use only and is not to be used in diagnostic or clinical procedures.

Scientific BackgroundThe matrix metalloproteinases (MMPs) are a large family of zinc endopeptidases. All MMPs are synthesized as inactive proenzymes. The activation of these proenzymes is a critical step that leads to degradation of extracellular matrix components such as fibronectin and collagen type III. MMP2 is activated by other MMPs such as MMP14 and MMP16. MMP2 is known to degrade collagen types IV, V, VII and X as well as gelatin type I.
