  • Scientific Background NAD-dependent protein deacetylas. Activates or deactivates mitochondrial target proteins by deacetylating key lysine residues. Known targets include ACSS1, IDH, GDH, SOD2, PDHA1, LCAD, SDHA and the ATP synthase subunit ATP5PO. Contributes to the regulation of the cellular energy metabolism (PubMed:24252090). Important for regulating tissue-specific ATP levels. In response to metabolic stress, deacetylates transcription factor FOXO3 and recruits FOXO3 and mitochondrial RNA polymerase POLRMT to mtDNA to promote mtDNA transcription. Acts as a regulator of ceramide metabolism by mediating deacetylation of ceramide synthases CERS1, CERS2 and CERS6, thereby increasing their activity and promoting mitochondrial ceramide accumulation

    Images, Protocols & SDS

  • A. Western Blot: (0.3 µg/mL) staining of Mouse Heart lysate (35 ug protein in RIPA buffer). Detected by chemiluminescence. B. Immunofluorescence analysis: of paraformaldehyde fixed NIH3T3 cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (2 µg/mL), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (2 µg/mL). C. Flow cytometry analysis: of paraformaldehyde fixed NIH3T3 cells (blue line), permeabilized with 0.5% Triton, showing a low level of staining. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (1 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody.
