Mouse monoclonal antibody to NeuN/Fox3 [1B7] (1-99): IgG

Description Fox3 is one of a family of mammalian homologues of Fox-1. The Fox proteins are about 46kDa in size, and each includes a central highly conserved RRM type RNA recognition motif. Much interest has focused on Fox3 as a result of the recent finding that this protein corresponds to NeuN, a neuronal nuclear antigen. NeuN/Fox-3 has a function in RNA splicing and is expressed heavily and specifically in neuronal nuclei and cytoplasm. Our antibody was raised against the N-terminal 100 amino acids of human Fox3 as expressed in and purified from E. coli. We did not use full length Fox3 as immunogen since the three mammalian Fox homologues, namely Fox1, Fox2 and Fox3, include virtually identical RRM motifs. The N-terminal region of the three molecules are much more variable in the three molecules so antibodies specific for each of the three molecules can therefore be generated.

Applications Western Blotting (WB), Immunohistochemistry (IHC) and Immunocytochemistry (IC). A dilution of 1:5,000 - 1:10,000 is recommended for WB. A dilution of 1:1,000 is recommended for IHC. A dilution of 1:500 - 1:1,000 is recommended for IC. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.
Specificity The specificity of this antibody has been confirmed by WB. Two alternate transcripts can be seen at 46 kDa and 48 kDa.

Mouse monoclonal antibody to NeuN/Fox3 detects expected doublet band for NeuN at ~46-48 kDa. Additional, uncharacterized bands are observed. Mouse monoclonal antibody to NeuN/Fox3 [1B7] (1-99): IgG Analysis of rat brain stem by Immunohistochemistry, co-stained with mouse antibody to Fox3/NeuN (M-377-100, green), and chicken antibody to microtubule associated protein 2 (MAP2, C-1382-50, red). Blue: DAPI staining of nuclear DNA. M-377-100 selectively stains nuclei and the proximal cytoplasm of neuronal cells, while the MAP2 antibody labels dendrites and overlaps with Fox3/NeuN staining in the perikarya of neurons. IHC Method: following transcardial perfusion with 4% paraformaldehyde, the brain was post fixed for 24 hours, cut to 45 um sections, and free-floating sections were stained with the two antibodies.
Mouse monoclonal antibody to NeuN/Fox3 [1B7] (1-99): IgG Rat brain neural cultures stained with Mouse monoclonal antibody to NeuN/Fox3 [1B7] M-377-100 (red), Chicken polyclonal antibody to GFAP C-1373-50 (green) and DNA (blue). The Fox3 antibody reveals strong nuclear and distal cytoplasmic staining for NeuN/Fox3 and the complete absence of staining of astrocytes, which are staining with the GFAP antibody, and other kinds of non-neuronal cells. This antibody is therefore an excellent marker of neuronal cells. Note that the same immunogen has been used to generate a rabbit polyclonal antibody to NeuN/Fox3 - see product R-3770-100.
Mouse monoclonal antibody to NeuN/Fox3 [1B7] (1-99): IgG Staining of NeuN and identification of neurons in fixed and permeabilized rat hippocampal cells by Immunocytochemistry. NeuN-IR (red) is localized to cell nuclei (blue) and appears pink due to overlaying colors.
