Rabbit polyclonal antibody to NeuN/Fox3 (5-24): affinity purified

$297.00USD

Catalogue No. R-3771-100
Description Fox3 is one of a family of mammalian homologues of Fox-1. The Fox proteins are about 46 kDa in size, and each includes a central highly conserved RRM type RNA recognition motif. Much interest has focused on Fox3 as a result of the recent finding that this protein corresponds to NeuN, a neuronal nuclear antigen. NeuN/Fox-3 has a function in RNA splicing and is expressed heavily and specifically in neuronal nuclei and cytoplasm.
Batch No. See product label
Unit size 100 ug
Antigen Peptide corresponding to amino acids 5-24 of human FOX3 coupled to KLH
Antigen Location N-terminus
Antibody Type Rabbit polyclonal
Other Names Feminizing locus on X; Fox-1; Fox3; NeuN
Accession A6NFN3 RFOX3_HUMAN;
Produced in Rabbit
Purity Affinity purified.
Applications Western Blotting (WB), Immunocytochemistry (ICC) and Immunohistochemistry (IHC). A dilution of 1:500 - 1:1,000 is recommended for WB. A dilution of 1:5,000 - 1:10,000 is recommended for ICC and IHC. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.
Specificity NeuN/FOX3. The specificity of this antibody has been confirmed by WB and IHC. Two alternate transcripts can be seen at 46 kDa and 48 kDa in WB. This antibody has been used successfully for immunohistochemistry on paraformaldehyde fixed rat brain (adult) tissue. It does not bind to the nuclei of perikarya of non-neuronal cells making it a good marker to identify neurons.
Species Against Human
Cross-reactivity Rat, mouse
Form Lyophilized from a solution containing PBS, pH 7.2-7.6, 0.1% trehalose, 5 mM sodium azide as preservative.
Reconstitution Reconstitute with 100 uL sterile-filtered, ultrapure water, to achieve an antibody concentration of 1 mg/mL. Centrifuge briefly to remove any insoluble material.
Storage Store lyophilized antibody at 2-8C. After reconstitution divide into aliquots and store at -20C for long-term storage. Store at 2-8C short-term (up to 4 weeks). Avoid repetitive freeze/thaw cycles.
Expiry Date 12 months after purchase

Rabbit polyclonal antibody to NeuN/Fox3 (5-24): affinity purified Left: Analysis of NeuN/FOX3 expression in adult rat brain section by Immunohistochemistry. Red: Rabbit anti-NeuN (1:2,000); Green: Chicken anti-MAP2 (C-1382-50, 1:5,000). IHC Method: Following transcardial perfusion with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45 um sections, and free-floating sections were stained. The FOX3/NeuN antibody selectively stains nuclei and cytoplasm of neuronal cells, while the MAP2 antibody labels dendrites and overlaps with Fox3/NeuN staining in neuronal perikarya. Right: Western blot analysis of cytosolic (cyt) and nuclear enriched (nuc) fractions of whole brain lysates (antibody dilution: 1:1,000). [1] MWM, [2] rat cyt, [3] rat nuc, [4] mouse cyt, and [5] mouse nuc lysate. Two bands of 46 kDa and 48 kDa correspond to the two alternate transcripts of the FOX3/NeuN protein. Western blotting was performed under non reducing conditions.
