- Application Details Immunohistochemistry (IHC) and Western Blotting (WB). A concentration of 2.0 µg/mL is recommended for WB. Human PCNA has a predicted length of 261 residues and MW of 29 kDa. A concentration of 0.4-1.0 µg/mL is recommended to detect PCNA in formalin fixed and paraffin embedded tissues as well as formalin/acetone fixed tissues. Antigen retrieval may improve staining. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.

PCNA helps increase the processivity of DNA polymerase during leading strand synthesis in DNA replication. In response to DNA damage, PCNA is ubiquitinated and is involved in the RAD6 dependent DNA repair pathway.
