Necdin is a growth suppressor that facilitates the entry of the cell into cell cycle arrest. It represses the activity of cell-cycle-promoting proteins such as SV40 large T antigen, adenovirus E1A, and the transcription factor E2F. Necdin also interacts with p53 and works in an additive manner to inhibit cell growth. Functions also as transcription factor and binds directly to specific guanosine-rich DNA sequences. Defecs in necdin may be the cause of the Prader-Willi syndrome which is characterised by muscular hypotonia, mental retardation, short stature, obesity, hypogonadotropic hypogonadism, and small hands and feet.
Immunohistochemical detection of Necdin on formalin fixed, paraffin-embedded, human colon. Anti-Necdin primary antibody, catalogue number M-847-100, was used at a concentration of 1 µg/mL.
Immunofluorescent detection of Necdin on HeLa cells. Mouse monoclonal antibody to human Necdin, catalogue number M-847-100, was used at a concentration of 10 µg/mL.
Western blot detection of Necdin expression in human promyelocytic leukemia cell lysate.
