    Images, Protocols & SDS

  • Left: Mixed neuron-glial cell cultures stained with anti-CNP (red) and anti-GFAP (green, R-1374-50) by Immunocytochemistry. The antibody to CNP stains strongly in oligodendrocytes, whereas GFAP labels only the intermediate filaments in astrocytes. Blue is DNA staining. Right: Western Blotting of rat brain tissue homogenates probed with anti-CNP antibody at 1:5,000 (Lane 1) and 1:20,000 (Lane 2). The antibody binds strongly and specifically to a band at ~48 kDa corresponding to the expected molecular weight of CNP.

  • Left: Rat cerebellum stained with mouse anti-CNP (green, 1:500) and chicken anti-NF-M (C-1393-50, red, 1:1,000) by Immunohistochemistry. Blue: DAPI nuclear stain. IHC Method: Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45 um, and free-floating sections were stained. The CNP antibody specifically stains oligodendrocytes, cells that create the myelin sheath around axons. The NF-M antibody labels axons of neuronal cells. Right: Western blot analysis of rat brain (2), rat spinal cord (3), mouse brain (4), mouse spinal cord (5). Antibody dilution: 1:2,000. Double bands at 46 and 48 kDa correspond to the major isotypes of CNP.
