Scientific BackgroundCell surface protein involved in cell-cell-interactions, exocytosis of secretory granules and regulation of signal transmission. Function is isoform-specific. Alpha-type isoforms have a long N-terminus with six laminin G-like domains and play an important role in synaptic signal transmission. Alpha-type isoforms play a role in the regulation of calcium channel activity and Ca2+-triggered neurotransmitter release at synapses and at neuromuscular junctions. They play an important role in Ca2+-triggered exocytosis of secretory granules in pituitary gland (Uniprot).
A. Western Blot: HEK293 lysate (10 ug protein in RIPA buffer) overexpressing Human NRXN1 with DYKDDDDK tag probed with G-1890-100 (0.5 µg/mL) in Lane A and probed with anti- DYKDDDDK Tag (1/3000) in lane C. Mock-transfected HEK293 probed with G-1890-100 (1 mg/mL) in Lane B. Detected by chemiluminescence. B. Flow cytometryanalysis: of paraformaldehyde fixed Kelly cells (blue line), permeabilized with 0.5% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (1 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody.
