Scientific BackgroundNFIL-3 is a protein coding gene that binds as a homodimer to activating transcription factor (ATF) sites in many cellular and viral promoters. The encoded protein represses PER1 and PER2 expression and therefore plays a role in the regulation of circadian rhythm. Three transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Feb 2014]
A. Western Blot: (1 µg/mL) staining of Jurkat nuclear cell lysate (RIPA buffer, 20 ug total protein per lane). Detected using chemiluminescence B. Flow cytometry analysis: paraformaldehyde fixed Human peripheral blood mononuclear cells (blue line), permeabilized with 0.5% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (2 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody.
