Scientific BackgroundActs as a transcriptional regulator that recognizes and binds to the sequence 5'-[GA]TTA[CT]GTAA[CT]-3', a sequence present in many cellular and viral promoters (Uniprot).
A. Flow cytometry analysis: paraformaldehyde fixed HepG2 cells (blue line), permeabilized with 0.5% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (0.4 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody. B. Immunofluorescence analysis: paraformaldehyde fixed HeLa cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (4 µg/mL), showing strong nuclear and some cytoplasmic staining. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (4 µg/mL). C. Western Blot: (0.3 µg/mL) staining of HepG2 (A) and negative control KLY (B) lysate (35 ug protein in RIPA buffer) Detected by chemiluminescence
A. Human Liver Cell Staining: (2 µg/mL) staining of paraffin embedded Human Liver. Steamed antigen retrieval with citrate buffer pH 6, HRP-staining. B. Immunofluorescence analysis: paraformaldehyde fixed HepG2 cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (4 µg/mL), showing strong nuclear and some cytoplasmic staining. The nuclear stain is DAPI (blue).
