Scientific BackgroundThe protein encoded by TRIM28 gene mediates transcriptional control by interaction with the Kruppel-associated box repression domain found in many transcription factors. The protein localizes to the nucleus and is thought to associate with specific chromatin regions. The protein is a member of the tripartite motif family. This tripartite motif includes three zinc-binding domains, a RING, a B-box type 1 and a B-box type 2, and a coiled-coil region. [provided by RefSeq, Jul 2008]
A. Flow cytometry analysis: paraformaldehyde fixed HeLa cells (blue line), permeabilized with 0.5% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (2 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody. B. Staining Human Breast Tissue: (2 µg/mL) staining of paraffin embedded Human Breast. Steamed antigen retrieval with Tris/EDTA buffer pH 9, HRP-staining.
A. Immunofluorescence analysis: paraformaldehyde fixed HeLa cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (2 µg/mL), showing nuclear staining. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10 µg/mL) followed by Alexa Fluor 488 secondary antibody (2 µg/mL). B.
