    Images, Protocols & SDS

  • Left: Mixed neuron-glial cell cultures stained with chicken anti-CNP (red) and mouse anti-GFAP antibody (M-1375-100) by Immunocytochemistry. The CNP antibody stains strongly in oligodendrocytes, whereas GFAP labels the intermediate filament in astrocytes. Blue: Nuclear DNA stain, which reveals the presence of neurons which do not express either CNP or GFAP. Right: Western Blot analysis of rat brain tissue homogenate with chicken anti-CNP antibody (1:5,000). One specific band at ~48 kDa is detected corresponding to the molecular weight of CNP.

  • Left: Rat cerebellum section stained with chicken anti-CNP (green, 1:2,000) and rabbit anti-NF-H antibody (red, 1:10,000). IHC Method: Following transcardial perfusion with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45 um, and free-floating sections were stained. The CNP antibody stains myelin and oligodendrocytes, cells that create the myelin sheath around axons. The NF-H antibody labels the heavily phosphorylated axonal forms of NF-H which are localized in large projection axons. Right: Western blot analysis of spinal cord tissue homogenates using chicken anti-CNP (red, 1:5,000). [2] mouse, [3] rat and [4] cow. Doublet bands at 46 and 48 kDa correspond to CNP isotypes. The blot was simultaneously probed with mouse anti-internexin antibody (green).
