Once in the nucleus, Smad proteins target a number of DNA binding proteins to stimulate transcription. Smad4, also known as the common-mediator Smad (co-Smad), interacts with the receptor-regulated Smad proteins (R-SMAD). The R-SMAD class includes Smad2 and Smad3.
