SpecificityThe specificity of this antibody has been confirmed by WB and IHC against the antigen. Human; predicted to react with mouse and rat due to sequence similarity.
Species ReactivityHuman, Mouse (Predicted), Rat (Predicted)
Immunogen DescriptionA synthetic peptide corresponding to a sequence of human PRKCA (153-182aa MDHTEKRGRIYLKAEVADEKLHVTVRDAKN), that differs from the related rat and mouse sequences by one amino acid. There is 100% alignment with various monkey species including Rhesus, Baboon, gorillas by BLAST as well as other mammals including horse and marmoset.
ConjugateUnconjugated
Purity DescriptionAffinity purified on antigen column
Application DetailsImmunohistochemistry (IHC) and Western Blotting (WB). A concentration of 0.1-0.5 µg/mL is recommended for WB. Human Protein kinase C alpha has a predicted length of 672 residues and MW of 77 kDa. A concentration of 0.5-1 µg/mL is recommended to detect the protein in paraffin embedded tissues. Antigen retrieval by boiling paraffin sections in 10 mM citrate buffer, pH 6.0 for 20 minutes is required. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.

Reconstitution InstructionsSpin vial briefly before opening. Reconstitute vial in 0.2 mL sterile-filtered, ultrapure water. This will produce a final concentration of 500 µg/mL of antibody. Centrifuge to remove any insoluble material.
Storage InstructionsAt least 12 months after purchase at -20°C (lyophilized formulations). After reconstitution, aliquot and store at -20°C for up to 6 months for higher stability.Avoid freeze-thaw cycles.
Batch NumberPlease see item label.
Expiration Date12 months after date of receipt (unopened vial).

Scientific BackgroundProtein kinase C alpha (PKC-alpha) belongs to the Protein kinase C family of serine- and threonine-specific protein kinases that can be activated by calcium and second messenger diacylglycerol. PKC-alpha is one of at least 12 different types of Protein kinase C identified in humans. PKC-alpha is ubiquitously expressed and has roles in many different cellular processes including cell adhesion, cell transformation, proliferation, differentiation and cell cycle checkpoint.
