Application DetailsImmunohistochemistry (IHC) and Western Blotting (WB). A concentration of 0.1-0.5 µg/mL is recommended for WB. Human NT3 has a predicted length of 257 amino acids and MW of 29 kDa. A concentration of 0.5-1.0 µg/mL is recommended to detect NT3 in paraffin embedded tissues. Antigen retrieval by boiling the paraffin / formalin section in 10mM citrate buffer, pH 6.0 for 20 minutes is required. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.

Scientific BackgroundNT3 is a member of the neurotrophin family, that controls survival and differentiation of visceral and proprioceptive sensory neurons. NT3 is closely related to both NGF and BDNF. It may be involved in the maintenance of the adult nervous system, and may affect development of neurons in the embryo when it is expressed in human placenta. NT3-deficient mice generated by gene targeting display severe movement defects of the limbs. The mature peptide of this protein is identical in all mammals examined including human, pig, rat and mouse. SUBCELLULAR LOCATION: Secreted protein. TISSUE SPECIFICITY: Brain and peripheral tissues. SIMILARITY: Belongs to the NGF-beta family.
Immunohistochemical staining of paraffin-embedded rat brain tissue sections using NT3 antibody (R-1016-100) at a concentration of 0.5-1.0 µg/mL. Secondary detection with Biotinylated goat anti-rabbit IgG (10 µg/mL).
