Product DescriptiongoogleThe rat TrkA Kit is a sandwich ELISA. The capture antibody is a polyclonal rat TrkA antibody pre-coated onto the 96-well strip plates provided in the kit. Test samples and standards of known TrkA concentration are added to these wells and allowed to complex with the bound TrkA antibody. A biotinylated rat polyclonal antibody is then added. This detection antibody binds to the antigen thus completing the sandwich. After washing, an enzyme Avidin-Biotin-Peroxidase complex (ABC) is added which binds to the second antibody. The peroxidase substrate TMB is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of TrkA present in the samples. The purpose of this kit is the in-vitro quantitative determination of rat TrkA in samples such as culture supernatant and cell lysates. This kit has been configured for research use only and is not to be used in diagnostic or clinical procedures.

Scientific BackgroundTrkA is a member of the neurotrophic tyrosine kinase receptor family. It is a membrane-bound receptor that upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. TrkA is required for high-affinity binding to nerve growth factor (NGF), neurotrophin-3 and neurotrophin-4/5 but not brain-derived neurotrophic factor (BDNF). TrkA leads to cell differentiations and may play a role in specifying sensory neuron subtypes. It has a crucial role in the development and function of the nociceptive reception system as well as establishment of thermal regulation via sweating.
