Left: Analysis of SH-SY5Y cells for UCHL1 expression with rabbit antibody to UCHL1 (green, 1:1,000) by Immunocytochemistry. Cells were co-stained with mouse antibody to fibrillarin (M-1372-250, red, 1:1,000). Blue: DAPI nuclear stain. The UCHL1 antibody produces strong staining of the cellular cytoplasm of these cells which share many properties with neurons, while the fibrillarin antibody specifically labels nucleoli. Right: Western blot analysis of tissue and cell lysates using rabbit antibody to UCHL1 (red, 1:2,000, lanes 2-7). protein standard, rat brain, mouse brain, NIH-3T3, HEK293, HeLa, SH-SY5Y cells. The single band at 24 kDa corresponds to the UCHL1 protein. UCHL1 is detectable in CNS extracts and cells with neuronal properties, but not in HeLa, NIH-3T3 and other non-neuronal cells. The blot was simultaneously probed with a mouse antibody to HSP60 (green).
