Application DetailsIHC: Use at an amount of 10 µg/mL with an incubation time of 1-3 days at 4ºC. This antiserum works in paraffin and 4% PFA fixed frozen sections. Penetration is the key to success. Over-fixed tissue is problematic. Not recommended for western blotting applications. Mouse monoclonal antibody M-1752-100 or rabbit polyclonal antibody R-1751-50 are excellent alternatives for western blotting. Biosensis recommends optimal dilutions/concentrations should be determined by the end user.

Reconstitution InstructionsSpin vial briefly before opening. Reconstitute in 500 µL sterile-filtered 1X PBS, pH 7.2-7.6. Centrifuge to remove any insoluble material.
Storage InstructionsAfter reconstitution keep aliquots at -20°C for a higher stability, and at 2-8°C with an appropriate antibacterial agent. Glycerol (1:1) may be added for an additional stability. Avoid repetitive freeze/thaw cycles.

Scientific BackgroundFUNCTION: Nuclear phosphoprotein which forms a tight but non-covalently linked complex with the JUN/AP-1 transcription factor. Has a critical function in regulating the development of cells destined to form and maintain the skeleton. It is thought to have an important role in signal transduction, cell proliferation and differentiation. SUBUNIT: Heterodimer. Interacts with DSIPI; this interaction inhibits the binding of active AP1 to its target DNA. Interacts with MAFB. SUBCELLULAR LOCATION: Nucleus. INDUCTION: C-fos expression increases upon a variety of stimuli, including growth factors, cytokines, neurotransmitters, polypeptide hormones, stress and cell injury. cFos was induced in the hypothalamic arcuate nucleus of an adult male Wistar rat using i.p. injection of N-methyl-D-aspartate. The brain was fixed by perfusion with formalin (4%). cFos immunoreactivity was detected in floated cryo-sections of the hypothalamic arcuate nucleus with the S-033-500 primary antibody (1:30 000) using the biotinylated secondary antibody-ABC method and nickel-diaminobenzidine chromogen.
