Reconstitution InstructionsSpin vial briefly before opening. Reconstitute in 100 µL sterile-filtered, ultrapure water. Centrifuge to remove any insoluble material.
Storage InstructionsMaintain lyophilized antibody at 2-8°C for up to 12 months after date of receipt. After reconstitution keep undiluted aliquots at -20°C for up to 6 months for higher stability or at 2-8°C with an appropriate antibacterial agent. Glycerol (1:1) may be added for additional stability. Avoid repetitive freeze/thaw cycles.

Scientific BackgroundNerve growth factor (NGF) is synthesized as a precursor (proNGF) which may be released and have physiological functions to cause cell death. It binds neurotrophin receptor p75 and sortilin and may also be important for the development of nervous system. proNGF is synthesized in target tissues and glia, transported retrogradely and may be released. This antibody is raised in sheep to detect the prodomain of NGF not the mature peptide.
Western blot of proNGF proteins produced in E.Coli. S-080-100 detects the human and mouse form of proNGF as well as the pro-domain peptide. SDS-PAGE: denatured and reduced; Transfer: Tris-Glycine buffer; Membrane: nitrocellulose (0.45 um); Blocking: 5% skim milk in TBST, 2 hours at RT; Primary antibody: overnight at 4°C (1:1000 dilution); Secondary antibody: anti-sheep-HRP (1/3000), 2 hours at RT; Detection: Chemiluminiscence.
Immunofluorescence analysis of proNGF expression in human DU145 prostate cancer cells. Fixed (4% formaldehyde), permeabilized, and blocked (10% normal horse serum, 0.1% Triton X100) DU145 cells were incubated with proNGF antibody S-080-100 (1:1000, red) for 1 hour. Primary antibody binding was visualized with a secondary donkey anti-sheep-PE antibody (1:100, 1 hour incubation). Cell nuclei were stained with Hoechst dye (blue). Magnification: 100x.
The sheep anti-proNGF antibodies recognize recombinant proNGF, the prodomain of NGF and but not mouse mature NGF (50 ng in each lane).
